Bonamia ostreae, parasite of the European flat oyster, Ostrea edulis, does not experimentally infect the Japanese oyster, Crassostrea gigas ArchiMer

The protozoan Bonamia ostreae (Ascetospora) is the etiological agent of bonamiosis, or hemocyte disease of the European fiat oyster, Ostrea edulis (Pichot et al., 1980). Since this infection was first reported from the coast of Brittany (Comps et al., 1980), the parasite has spread rapidly along the French Atlantic coasts (Grizel & Tigé, 1982), and in the European countries including the UK, Holland, lreland and Spain (Bannister & Key, 1982; Van Banning, 1982; Polanco et al., 1984). Under the EEC Council Directive 91/67, this parasitic protozoan is included in list II of Annex A. Serious pathogen agents for molluscs that should be obligatorily declared are placed in this list

Les mortalités d'huîtres observées sur les côtes françaises apparaissent principalement à proximité du sédiment et pendant la gamétogenèse. Les interactions de facteurs environnementaux sur les écosystèmes conchylicoles comme la température, les apports des bassins versants et le compartiment sédimentaire illustrent un risque de mortalité lié à différentes sources de stress (Thème 1). Cellular and molecular responses of haemocytes from Ostrea edulis during in vitro infection by the parasite Bonamia ostreae ArchiMer

Bonamia ostreae is a protozoan, affiliated to the order Haplosporidia and to the phylum Cercozoa. This parasite is intracellular and infects haemocytes, cells notably involved in oyster defence mechanisms. Bonamiosis due to the parasite B. ostreae is a disease affecting the flat oyster, Ostrea edulis. The strategies used by protozoan parasites to circumvent host defence mechanisms remain largely unknown in marine bivalve molluscs. Characterization of abalone Haliotis tuberculata-Vibrio harveyi interactions in gill primary cultures ArchiMer

The decline of European abalone Haliotis tuberculata populations has been associated with various pathogens including bacteria of the genus Vibrio. Following the summer mortality outbreaks reported in France between 1998 and 2000, Vibrio harveyi strains were isolated from moribund abalones, allowing in vivo and in vitro studies on the interactions between abalone H. tuberculata and V. harveyi. This work reports the development of primary cell cultures from abalone gill tissue, a target tissue for bacterial colonisation, and their use for in vitro study of host cell—V. harveyi interactions. Characterization of summer mortalities of C. gigas oyster in France in relation to environmental parameters ArchiMer

Field characterization of summer mortality was performed in France in the frame of the Morest project. Natural and hatchery spat were compared between three oyster production areas in France. Regardless of the natural or hatchery origin, oysters died during the reproduction period after temperature reaches 19 degree C. Thus, in southern areas, temperature accelerated gametogenesis of small spat (10mm) as well as adults, and mortality appeared for the two age classes. In contrast, sexual maturation proceeded more slowly in northern where spat mortality was lower compared to 18 months old oysters. Characterization of Vibrio isolated from Crassostrea gigas spat suffering summer mortality outbreaks ArchiMer

The French oyster production is mainly based on the pacific oyster, Crassos/rea gigas. However, for several years, C. gigas spat has suffered severe mortality outbreaks. A multifactorial etiology implicating environment, oyster physiology, genetics and pathogens is heavily suspected. A herpesvims and bacteria belonging to the Vihrio s\p/endidus polyphyletic group have been isolated From summer mortality outbreaks. Moreover, several strains belonging to the genus Vibrio have been identified as pathogen for different mollusk species. However, few tools are available for the identification of these Vibrios and for the characterization of the virulence of the pathogenic strains.

The pathogens related to summer mortality outbreaks are a herpes virus and two bacterial strains one belonging to Vibrio splendidus biovar II and the other to Vibrio splendidus spp. However, the feature pathogen/opportunist of these strains is still unknown. Several strains belonging to the genus Vibrio have been identified as pathogen for different mollusk species. In the context of the French program Morest, experiments of cohabitation have been used to demonstrate the potential presence of a transmissible infectious agent in batches of oysters suffering from summer mortality outbreaks. Chlamydia-like organisms in ctenidia and mantle cells of the Japanese oyster Crassostrea ggias from the French Atlantic coast ArchiMer

Grossly visible lesions of the ctenidia and occasional mortalities in Japanese oyster Crassostrea gigas from the Atlantic French coast were reported in 1992-1993. From August 1992 to March 1993, 235 living oysters were collected to determine the cause of tissue lesions and mortalities. Microscopical examination of fixed, stained sections revealed that 53 2% of these oysters had ctenidia and mantle lesions associated with irregular basophllic and Feulgen positive cytoplasmic inclusion bodies. Ultrastructurally, the infectious agent was a chlamydia-like organism. Combination of a pesticide exposure and a bacterial challenge: In vivo effects on immune response of Pacific oyster, Crassostrea gigas (Thunberg) ArchiMer

To assess the impact of pollution induced by pesticides on Pacific oyster, Crassostrea gigas, health in France, in vivo effects of combined pesticide exposure and bacterial challenge on cell activities and gene expression in hemocytes were tested using flow cytometry and real-time PCR. As a first step, an in vivo model of experimental contamination was developed. Pacific oysters were exposed to a mixture of eight pesticides (atrazine, glyphosate, alachlor, metolachlor, fosetyl-alumimium, terbuthylazine, diuron and carbaryl) at environmentally relevant concentrations over a 7-day period. Massive mortality outbreaks affecting Pacific oysters (Crassostrea gigas) spat/juveniles are often associated with the detection of a herpesvirus called ostreid herpesvirus type 1 (OsHV-1). In this work, experimental infection trials of C. gigas spat with OsHV-1 were conducted using two contrasted Pacific oyster families for their susceptibility to viral infection. Recent diagnostic successes show that marine disease diagnosis requires both modern, cutting-edge technology (e.g. metagenomics, quantitative realtime PCR) and more classic methods (e.g. transect surveys, histopathology and cell culture). Here, we discuss how this combination of traditional and modern approaches is necessary for rapid and accurate identification of marine diseases, and emphasize how sole reliance on any one technology or technique may lead disease investigations astray. Complete genome sequence of acute viral necrosis virus associated with massive mortality outbreaks in the Chinese scallop, Chlamys farreri ArchiMer

Acute viral necrosis virus (AVNV) is the causative agent of a serious disease resulting in high mortality in cultured Chinese scallops, Chlamys farreri. We have sequenced and analyzed the complete genome of AVNV. The AVNV genome is a linear, double-stranded DNA molecule of 210,993 bp with a nucleotide composition of 38.5% G + C. A total of 123 open reading frames were predicted to encode functional proteins, ranging from 41 to 1,878 amino acid residues. The DNA sequence of AVNV is 97% identical to that of ostreid herpesvirus 1 (OsHV-1), and the amino acid sequences of the encoded proteins of these two viruses are 94-100% identical. Compte-rendu des résultats d'analyses effectuées en histologie - Année 1995 ArchiMer

Dans le cadre du Réseau zoosanitaires CEE (huître plate) et suivi d'autres espèces, 890 individus provenant des zones 5, 6 et 7 ont été examinés sur l'ensemble de l'année 1995 par différentes techniques : 472 analyses sous coupes histologiques, 328 analyses sur appositions d'organes, 90 analyses sur milieu au thioglycolate.
